In December I arranged a tourist visa for 2 months through a courier, which cost 7000 baht, in Cambodia. But want a new tourist visa again in February.
Do I have to go back to Cambodia or can I just go to the Thai immigration office here in Sisaket?

You can extend a tourist visa for 1900 baht for 30 days at any immigration office in Thailand.
For a new tourist visa you will have to go to an embassy or consulate and they are only located outside Thailand, so yes you will have to go to Cambodia, Laos (where they also issue double entries), Malaysia or something.
It depends on how long you want to stay after your current visa expires and whether you can meet certain conditions, otherwise these are options. (See also Visa file)
1. If you want to stay for a short period, you can request an extension of 30 days on your TR Visa, possibly supplemented with a 15-day visa run to the border, or 30 days via a cheap flight that takes you outside Thailand and possibly the same day brings back. Might cost you less than 7000 Bath.
2. If you want to stay longer, you can, if you meet the conditions, have your TR visa converted into an O visa (Multiple entry possible?) at immigration.
I do not know your age and details, but perhaps a one-year extension is also possible after obtaining the O visa

If necessary, go to Pattaya Immigration where they are more familiar with applications such as visa conversions.
3. Get a new TR visa outside Thailand.
Have a visa extended with a courier via Cambodia!! It smells very bad indeed.
The passport is a personal travel document, using the passport in this way is not wise in my opinion.
If that courier is caught, you will also lose the hare and your passport. Then you have some explaining to do at the Ned Embassy and at Thai immigration.
My gut feeling says never again.
Geert, I hope for your sake that the stamp your courier received upon entering Thailand is a real stamp. There is a chance that this stamp has been counterfeited. If immigration sees this, then you are the cigar for just saying so. Never do this again!

1. You can normally get your TR visa in the neighboring countries you mention.
Keep in mind that if you repeatedly and consecutively obtain TR visas, people will sometimes ask whether you do not want to stay in Thailand for a long time and your purpose is other than tourism. Refusal can be a consequence.
2. If by annual visa, you mean a Non-Immigrant “O” Multiple Entry then it is yes.

Do not leave Thailand anymore, of course, because your visa has been used up.
